囊泡亚群蛋白解析芯片用于退行性疾病诊断

Parkinson’s disease (PD) is characterized neuropathologically by α-synuclein aggregation. Currently, there is no blood test that can predict the underlying pathology or offer early diagnosis in the prodromal phase of the disease. We investigated the potential clinical utility of serum brain-derived extracellular vesicles (BDEVs) as biomarkers. Due to the low abundance of BDEVs subpopulations and the complexity of the sample matrix, we developed (1) antifouling immunobeads to improve specific EV immunocapture, (2) stimuli-responsive isolation platforms for intact and active EV capture and release, (3) digital droplet microfluidic assay to evaluate the EV membrane antigens on single-vesicle level. Such platforms have established a promising PD biomarker based on neuronal marker measurements and offer the potential for the molecular stratification of patients with parkinsonism.
